BACKGROUD: Macrobrachium rosenbergii is an economically important species that is widely cultivated in some Asian nations. Foxl2 is a transcriptional regulator of ovarian differentiation and development. The aim of this study was to study the bioinformatics features and expression patterns of M. rosenbergii Foxl2 (MrFoxl2). METHODS: In this study, all experimental animals were mature M. rosenbergii (9-12 cm) individuals. The foxl2 gene was identified and characterized in the genome of M. rosenbergii using molecular cloning, bioinformatic analysis, in situ hybridization, and quantitative analysis. RESULTS: The identified cDNA encoded a putative 489-amino-acid MrFoxl2 protein. Bioinformatics analysis revealed a low identity of MrFoxl2 to other crustacean orthologues. The closest phylogenetic relationship was to Foxl2 of Eriocheir sinensis. The result of in situ hybridization demonstrated that transcripts of MrFoxl2 in M. rosenbergii were identified in spermatocytes, oocytes, and secretory epithelial cells of the vas deferens. The result of q-PCR suggested that a high expression of MrFoxl2 was identified in the testis, vas deferens, and ovaries. During ovarian development, MrFoxl2 expression was the highest in stage I. CONCLUSION: Our findings suggest that MrFoxl2 may play a role in gonadal development in both female and male M. rosenbergii.

BACKGROUND: Cold stress is one of the abiotic stresses that affect plant growth and development, as well as life and geographical distribution important. For researching how plants react to low temperature stress, Rhododendron chrysanthum Pall. (R. chrysanthum) growing in Changbai Mountains of China is an essential study subject. METHODS AND RESULTS: R. chrysanthum was cold-treated at 4 °C for 12 h (cold-stress group-CS, and controls-CK), combined with transcriptomics (RNA-seq) and proteomics (iTRAQ) techniques, to investigate the response mechanisms of R. chrysanthum response to cold stress. Cold stress resulted in the discovery of 12,261 differentially expressed genes (DEGs) and 360 differentially expressed proteins (DEPs). Correlation of proteomic and transcriptome data, proteome regulation of distinct subcellular localization, and gene/protein functional groupings are all part of the investigation. CONCLUSIONS: The combined analysis showed that 6378 DEPs matched the corresponding DEGs when the control was compared with the cold-treated samples (CK vs CS). The analysis identified 54 DEGs-DEPs associated with cold stress. cold-tolerant DEGs-DEPs were enriched with hydrolase activity, acting on glycosyl bonds, carbon-oxygen lyase activity and ferric iron binding. Seven potential DEGs-DEPs with significant involvement in the cold stress response were identified by co-expression network analysis. These findings identify the synergistic effect of DEGs-DEPs as the key to improve the cold tolerance of R. chrysanthum and provide a theoretical basis for further studies on its cold resistance subsequently.

BACKGROUND: The growth and yield of pepper (Capsicum annuum L.) is often affected by the critical salt stress. Salicylic acid (SA) can improve plants' stress tolerance by promoting growth and regulating ion absorption and transportation. METHODS AND RESULTS: To uncover the alleviated mechanism of salt stress by SA in pepper, we conducted morphological, physiological, cytological, and transcriptomic analyses under a single SA treatment and NaCl with and without SA pre-treatment for 9 days. Seedlings under NaCl treatment showed yellow shrunken leaves, this tatus were alleviated by NS treatment (NaCl with SA pre-treatment). Compared with plants under NaCl treatment, those in the NS treatment showed reduced lipid peroxidation, and significantly increased contents of chlorophyll and osmotic regulators (proline, soluble sugars). Treatment with SA balanced the Na+/K+ ratio. We conducted transcriptome sequencing and identified differentially expressed genes (DEGs) contributing to alleviation of salt stress by SA in pepper. Besides photosynthesis related genes, GO and KEGG analyses revealed that the DEGs were enriched in 'sequence-specific DNA binding', 'transcription regulator activity' and 'DNA binding transcription factor activity' by GO terms. And our results showed that TFs, such as MYB, bZIP, BBX, AP2/ERF, NAC, etc., probably make a great contribution in the alleviation of salt stress by SA. CONCLUSIONS: These results reveal that SA can improve plants' stress tolerance by balancing ion absorption, gene expression and transcriptional regulation, which provide new ideas and resources for subsequent research on the mechanism of salt tolerance in pepper.
